Reading Between the Lines of Qualitative Data – How to Detect Hidden Structure Based on Codes
While qualitative research is experiencing broad acceptance in the information systems discipline, growing volumes of heterogeneous data pose challenges to manual qualitative analysis. We introduce an unsupervised machine learning approach based on graph partitioning to detect hidden information and structure in qualitative data samples. With the clustering technique, we map coded data to a graph and formulate a partitioning problem which is solved by integer linear programming. As a result, clusters of information sources are identified based on similarities given in the coded data. We demonstrate the approaches’ ability to detect hidden information in coded qualitative data by application on coded interview transcripts. With the approach, we draw on a technique from the operations research discipline and expand the repertoire of approaches being used to analyze qualitative data in the context of information systems

"The panel study 'Labour Market and Social Security' (PASS), established by the Institute for Employment Research (IAB), is a new dataset for labour market, welfare state and poverty research in Germany, creating a new empirical basis for the scientific community and for policy advice. This Datenreport provides an overview of the second survey wave, for which 12,487 individuals were interviewed in 8,429 households between December 2007 and July 2008. 10,114 individuals and 7,342 households were interviewed for the second time in the context of PASS. The spectrum of questions and the design of PASS are intended to close gaps in the existing stock of data. PASS has three main characteristics that extend analysis potential beyond that of the Federal Employment Agency's administrative data: 1. The panel takes the household context into account - including the situation before and after receipt of Unemployment Benefit II. 2. The panel is complete in that it covers all groups of persons and all employment biographies, not only people in dependent employment, unemployed people and those in need of assistance. The dataset also provides information on the status during phases of economic inactivity, self-employment or employment as civil servants. 3. Rare variants in NR2F2 cause congenital heart defects in humans
Congenital heart defects (CHDs) are the most common birth defect worldwide and are a leading cause of neonatal mortality. Nonsyndromic atrioventricular septal defects (AVSDs) are an important subtype of CHDs for which the genetic architecture is poorly understood. We performed exome sequencing in 13 parent-offspring trios and 112 unrelated individuals with nonsyndromic AVSDs and identified five rare missense variants (two of which arose de novo) in the highly conserved gene NR2F2, a very significant enrichment (p = 7.7 × 10?7) compared to 5,194 control subjects. We identified three additional CHD-affected families with other variants in NR2F2 including a de novo balanced chromosomal translocation, a de novo substitution disrupting a splice donor site, and a 3 bp duplication that cosegregated in a multiplex family. NR2F2 encodes a pleiotropic developmental transcription factor, and decreased dosage of NR2F2 in mice has been shown to result in abnormal development of atrioventricular septa. Via luciferase assays, we showed that all six coding sequence variants observed in individuals significantly alter the activity of NR2F2 on target promoters
Terrorist incidents: strategic treatment objectives, tactical diagnostic procedures and the estimated need of blood and clotting products
BACKGROUND
Terrorism-related incidents that are associated with mass casualties (mass-casualty terrorist incidents) are a medical and organisational challenge for every hospital because of the special injury patterns involved, the time of the incident, the development of the situation, the initial lack of information, the number of injured, and the number of uninjured survivors who self-refer to a hospital.
METHODS
The Terror and Disaster Surgical Care (TDSC®) - Course was developed in order to address mass-casualty terrorist incidents and to provide surgeons with the specialist medical and surgical knowledge and skills required for these special situations. The focus of the TDSC® course is on how to provide surgical care and how to deploy scarce resources in a particular tactical situation in such a way that the number of survivors is maximised.
RESULTS
The effective management of such a tactical situation must be based on priorities and first and foremost requires the standardised sorting and categorisation of the injured at the hospital. The aim of triage, or the sorting of the injured, is to immediately identify patients with life-threatening injuries in environments with strained resources. The medical management of mass-casualty terrorist incidents requires tactical abbreviated surgical care (TASC) teams that have the skills needed to perform a primary survey and to provide care for casualties who need immediate surgery (triage category 1-T1). Initial fluid therapy should be restrictive (permissive hypotension) unless contraindicated. Clotting products are replaced in a standardised manner on the basis of patient requirements, which are calculated using rapidly available surrogates (blood gas analysis). Blood products can be administered or kept available depending on risks and triage categories. The highest priority should be given to the identification and management of haemodynamically unstable patients who require immediate surgery for injuries associated with bleeding into body cavities (T1 + +).
CONCLUSION
The recommendations and approaches described here should be considered as proposals for hospitals to develop standards or modify well-established standards that enable them to prepare themselves successfully for situations (e.g. mass-casualty terrorist or shooter incidents) in which their resources are temporarily overwhelmed
Photon upconverting nanoparticles for luminescent sensing of temperature
Photon upconverting nanoparticles convert near-infrared into visible light (anti-Stokes emission), which strongly reduces the background of autofluorescence and light scattering in biological materials. Hexagonal NaYF4 nanocrystals doped with Yb3+ as the sensitizer and Er3+/Ho3+/Tm3+ as the activator display at least two emission lines that respond differently to temperature changes. The ratio of the main emission line intensities enables a self-referenced optical readout of the temperature in the physiologically relevant range from 20 to 45 °C. Upconverting nanoparticles of the type NaYF4:Yb, Er covered by an inactive shell of NaYF4 are bright and allow for resolving temperature differences of less than 0.5 °C in the physiological range. HAX1 deletion impairs BCR internalization and leads to delayed BCR-mediated apoptosis
Deletion of HAX1 in mice causes a severe reduction in the numbers of lymphocytes in the bone marrow and in the spleen. Additionally, B220+ B progenitor cells in the bone marrow are reduced, suggesting an important function of HAX1 in B cell development. HAX1 is thought to play a protective role in apoptotic processes; therefore, we investigated the role of HAX1 in bone marrow B progenitor cells and splenic B cells. We did not observe an effect on the survival of Hax1/ bone marrow cells but detected enhanced survival of splenic Hax1/ B cells upon in vitro starvation/growth-factor withdrawal. To explain this apparent inconsistency with previous reports of HAX1 function, we also studied the B cell receptor (BCR)-induced apoptosis of IgM-stimulated splenic naïve B cells and found that apoptosis decreased in these cells. We further found impaired internalization of the BCR from Hax1/ splenic B cells after IgM crosslinking; this impaired internalization may result in decreased BCR signaling and, consequently, decreased BCR-mediated apoptosis. We measured HAX1 binding to the cytoplasmic domains of different Ig subtypes and identified KVKWI(V)F as the putative binding motif for HAX1 within the cytoplasmic domains. Because this motif can be found in almost all Ig subtypes, it is likely that HAX1 plays a general role in BCR-mediated internalization events and BCR-mediated apoptosis.(VLID)240564
